1 Scope
This standard is applicable to 1,3-dioleoyl-2-palmitoyl triglyceride, a kind of food nutritive
fortifier with edible vegetable oil and oleic acid (derived from edible vegetable oils and fats) as
raw material and prepared via conversion of lipase catalytic ester for food industry. For
preservation purpose, antioxidant may be contained in 1,3-dioleoyl-2-palmitoyl triglyceride.

2.1 Chemical name
1,3-dioleoyl-2-palmitoyl triglyceride

859.39 (in accordance with international relative atomic mass in 2007)
Triglyceride base group with same carbon number can be separated from the oil or fat solution via
gas chromatography under program-controlled temperature; then it can be confirmed by
comparing with the standard triglyceride solution. Carry out quantitative determination by internal
standard method, its content is calculated by the triglyceride with carbon number of C52.
A.3.2 Reagents and materials
A.3.2.1 n-hexane: chromatographically pure.
A.3.2.2 Standard product of glyceryl triheptadecanoate: purity ≥ 99%.
A.3.2.3 Standard product of 1,3-dioleoyl-2-palmitoyl triglyceride: purity ≥ 99%.
A.3.2.4 Standard product of palmitoyl triglyceride: purity ≥ 99%.
A.3.2.5 Nitrogen: purity ≥ 99.999%.
A.3.3 Apparatus
Gas chromatograph: equipped with hydrogen flame ionization detector (FID).
A.3.4 Referenced chromatographic condition
A.3.4.1 Chromatographic column: 100% dimethyl polysiloxane high-temperature resistant
chromatographic column (15m in length, 0.25mm in inner diameter and 0.1μm in film thickness)
or the chromatographic column with equivalent performance.
A.3.4.2 Sample inlet temperature: 320℃.
A.3.4.3 Detector temperature: 370℃.
A.3.4.4 Temperature programming: 300℃, keep for 3.0min, and rise to 380℃ at the rate of
20℃/min and keep for 10min.
A.3.4.5 Carrier gas: nitrogen, constant voltage mode.
A.3.4.6 Sample injection volume: 1.0μL.
A.3.4.7 Degree of separation shall be greater than 1.5.
A.3.5 Analytical procedures
A.3.5.1 Preparation of standard solution
A.3.5.1.1 Standard solution of glyceryl triheptadecanoate (2mg/mL): put 100mg standard
product of glyceryl triheptadecanoate in a 50mL volumetric flask, add into a proper amount of
n-hexane and dissolve it, then dilute to the scale and shake the mixture well.
A.3.5.1.2 Standard solution of 1,3-dioleoyl-2-palmitoyl triglyceride (1mg/mL): put 100mg of
standard product of 1,3-dioleoyl-2-palmitoyl triglyceride in a 100mL volumetric flask, add a
proper amount of n-hexane and dissolve it, then dilute to the scale and shake the mixture well.
A.3.5.1.3 Standard solution of palmitoyl triglyceride (1mg/mL): put 100mg standard product of
palmitoyl triglyceride in a 100mL volumetric flask, add into proper amount of n-hexane and
dissolve it, then dilute to the scale and shake the mixture well.
A.3.5.1.4 Preparation of mixed standard solution (0.2mg/mL): transfer 1.0mL of standard
solution of glyceryl triheptadecanoate, 2.0mL standard solution of 1,3-dioleoyl-2-palmitoyl
triglyceride and 2.0mL of standard solution of palmitoyl triglyceride into a 10mL volumetric flask,
add into a proper amount of n-hexane and dilute to the scale, then shake the mixture well. Typical
chromatogram of mixed standard solution and retention time of all components are detailed in
Annex B.
A.3.5.2 Preparation of specimen solution
Put 20mg of specimen into a 25mL volumetric flask, then add into a small amount of n-hexane,
dissolve the specimen and shake the mixture well. Add into 2mL of internal standard solution of
glyceryl triheptadecanoate. Use n-hexane to bring the volume to the scale and shake well.
A.3.5.3 Determination
Under the specified chromatographic condition, take 1.0μL of standard solution and 1.0μL of
specimen solution and inject them into the chromatograph, then record the peak area of
component in standard solution and peak area of specimen solution.
A.3.6 Result calculation
A.3.6.1 The mass fraction (wi) of the content of 1,3-dioleoyl-2-palmitoyl triglyceride and
palmitoyl triglyceride in the specimen shall be calculated according to Formula (A.2):

Ai — the peak area of 1,3-dioleoyl-2-palmitoyl triglyceride or palmitoyl triglyceride in specimen
solution;
AR — the peak area of glyceryl triheptadecanoate in chromatogram of specimen solution;
